Each plate was normalized using the Robust Z score method [14], using the DMSO negative control as reference, and single point data visualized as in Figure 4. As primary screening was performed in duplicate, hits were classified as described in Figure 4. Tier 1 through 3 hits (Prio 1) were hits where both repeats had a consistent and reproducible signal (Figure 5A). An additional 81 compounds (Tier 4/Prio 2) showed activity in one repeat alone, which were also taken forward for dose response testing. Figure 4. n=2 Single Point Screen Summar y Plot.
