These include: • Sensitivity: The technology must be sensitive enough to detect weak affinities with a high signal- to-noise ratio, ensuring accurate readings even at low protein concentrations. • In -Solution assays: Avoiding the immobilization of proteins minimizes artifacts and allows for high compound concentrations. Solution-based assays provide more accurate and consistent results when working in complex conditions. • Throughput: While screening millions of compounds may not be necessary, medium to high throughput is essential for processing hundreds or thousands of hits efficiently, particularly in hit characterization. • Minimal Protein Labeling: Technologies that don't require labeling of all involved proteins are preferable, as they are less prone to artifacts and therefore provide more reliable results, being closer to the native system.
