Evaluation of slow equilibration kinetics in antibody chemical denaturation experiments

This was observed for three distinct mAbs, suggesting that the very slow equilibration kinetics observed for partially unfolded mAbs might be a universal property of mAbs. Based on these results, we recommend to use long incubation times (overnight, at least 16 h) after preparation of the chemical denaturation samples. The results also show that a just-in-time preparation of samples prior to measuring chemical denaturation produces results which do not reflect the true equilibrium state of the system and are thus not suited to derive ΔG values. It is therefore advisable to de-couple sample preparation and measurement for mAb chemical denaturation experiments.
